The transcription factor 7-like 2 gene (TCF7L2) rs7903146 T allele is constantly associated with Type 2 diabetes in various populations and ethnic groups. Nevertheless, this has not been observed in two studies involving Arab populations. The aim of the present study was to investigate the association between TCF7L2 rs7903146 in an Iranian population. Type 2 diabetes patients (N = 258) and normal healthy control subjects (N = 168) from the same area, were examined. The ARMS-PCR (Amplification Refractory Mutation System) technique, subsequently validated by direct sequencing, was used for genotyping. Allele and genotype frequencies were significantly different between patients and controls TT vs. CT + CC [p 0.0081 OR 3.4 95%CI (1.27-11.9)] and T vs. C...

Transformação de plantas. Infoteca-e
ANDRADE, S. R. M. de.
ABSTRACT: During centuries mankind plants to obtain cultivars with superior characteristics. However, conventional methods of plant breeding present limitations. The development of transformation methods created possibilities of great progresses in plant breeding because it offers the opportunity to introduce exogenous genes into superior cultivars to obtain new varieties. Genetic transformation is the targed organism using non sexual methods. Transgenic plants can be defined as those the present genes different from the original genoma obtained by techniques of genetic engineering. The transformation stages consist of the identification isolation, cloning and introduction of gene into the cells. There are several transformation methods, and the choice...

Clinical and molecuar characterization of Brazilian patients with growth hormone gene deletions BJMBR
Arnhold,I.J.P.; Osorio,M.G.F.; Oliveira,S.B.; Estefan,V.; Kamijo,T.; Krishnamani,M.R.S.; Cogan,J.D.; Phillips III,J.A.; Mendonça,B.B..
Genomic DNA from 23 patients with isolated growth hormone (GH) deficiency (12 males and 11 females: heights -4.9 ± 1.4 SDS) was screened for GH gene deletions by restriction endonuclease analysis of polymerase chain reaction amplification products. Three unrelated patients had typical features of severe GH deficiency and deletions (6.7 kb in two and 7.6 kb in one) of the GH gene. The two patients with 6.7-kb deletions developed growth-attenuating anti-GH antibodies whereas the patient with the 7.6-kb deletion continued to grow with GH replacement therapy. Our finding that 3/23 (~13%) Brazilian subjects had GH gene deletions agrees with previous studies of severe isolated GH deficiency subjects in other populations. Two of three subjects (67%) with...

Molecular cloning and seasonal expression of oyster glycogen phosphorylase and glycogen synthase genes ArchiMer
Bacca, Helene; Huvet, Arnaud; Fabioux, Caroline; Daniel, Jean-yves; Delaporte, Maryse; Pouvreau, Stephane; Van Wormhoudt, A; Moal, Jeanne.
To investigate the control at the mRNA level of glycogen metabolism in the cupped oyster Crassostrea gigas, we report in the present paper the cloning and characterization of glycogen phosphorylase and synthase cDNAs (Cg-GPH and Cg-GYS, respectively, transcripts of main enzymes for glycogen use and storage), and their first expression profiles depending on oyster tissues and seasons. A strong expression of both genes was observed in the labial palps and the gonad in accordance with specific cells located in both tissues and ability to store glucose. Cg-GPH expression was also found mainly in muscle suggesting ability to use glycogen as readily available glucose to. supply its activity. For seasonal examinations, expression of Cg-GYS and Cg-GPH genes...

Relationship between grain colour and preharvest sprouting-resistance in wheat PAB
Bassoi,Manoel Carlos; Flintham,John.
Since red alleles (R) of the genes that control grain colour are important for the improvement of preharvest sprouting resistance in wheat and there are three independently inherited loci, on chromosomes 3A, 3B and 3D of hexaploid wheat, it is possible to vary the dosage of dominant alleles in a breeding program. The objective of this work was to evaluate the dosage effect of R genes on preharvest sprouting, in a single seed descent population, named TRL, derived from the cross between Timgalen, white-grained wheat, and RL 4137, red-grained wheat. The study was carried out using sprouting data in ripe ears obtained under artificial conditions in a rainfall simulator over three years. According to the results there is a significant effect on preharvest...

Phenotypic and genotypic resistance profile of Salmonella Typhimurium to antimicrobials commonly used in poultry Rev. Bras. Ciênc. Avic.
Biffi,CP; Stefani,LM; Miletti,LC; Matiello,CA; Backes,RG; Almeida,JM; Neves,GB.
Isolates of Salmonella sp (104) from poultry samples were isolated and serotyped where eleven were identified as Salmonella Typhimurium (ST). ST isolates were phenotypically tested by disk diffusion and minimum inhibitory concentration (MIC). Four genes related to enrofloxacin (GyrA, GyrB, ParC and ParE), two to gentamicin (AadA and AadB) and two to ceftiofur (BlaCMY-2 and AmpC) resistance were searched by PCR. Our results showed ST resistance to all three antibiotics tested (18.1% for ceftiofur, 45.4% for gentamicin, and 18.1% for enrofloxacin) according to the diffusion test. In the MIC test, the ST isolates showed higher levels of resistance (27.2% for ceftiofur, 54.5% for gentamicin, and 18.2% for enrofloxacin). Three resistance genes out of four...

Development of a polymerase chain reaction and its comparison with agar gel immunodiffusion test in the detection of bovine leukemia virus infection Braz. J. Vet. Res. Anim. Sci.
Camargos,Marcelo Fernandes; Stancek,Daniel; Lessa,Leandro Moreira; Reis,Jenner Karlisson Pimenta; Rocha,Maurílio Andrade; Leite,Rômulo Cerqueira.
Polymerase chain reaction (PCR) was used for bovine leukemia virus (BLV) detection in the peripheral leukocytes of the infected bovines. The primers used were designed to amplify a part of env gene of BLV. PCR products were analyzed by agarose gel electrophoresis stained by ethidium bromide. The analytical specificity of PCR was confirmed by enzymatic restriction analysis of the PCR product with Bam HI and also by nucleotide sequence analysis of three PCR samples. Sixty five animals were tested for anti-BLV antibody, by agar gel-immunodiffusion test (AGID) and for direct BLV detection by PCR. There was a 73.80% concordance rate between the two tests. Four animals positive in AGID were PCR negative, while 13 AGID negative animals were found PCR positive....

Development of mussel-specific expression vectors suitable for transgenic organisms: implication on the establishment of mollusc continuous cell lines ArchiMer
Delsert, Claude; Cancela Da Fonseca, Leonor.
The main objective of this project was to develop the tools for the in-depth understanding of mollusc biology and pathology. Such a goal has been reached in other biological systems through the development of molecular biology and cell culture. Indeed, development of a mollusc cell culture system is an essential step to approach many of the problems related to mollusc pathology, in particular to use molecular biology tools to study viral infection of these organisms. No continous cell line is available from bivalves and while cell cultures have been obtained from these organisms they are difficult to develop and labor intensive, survival cultures being maintained only for a few weeks. Our goal has been to study in detail many of the parameters involved in...

Reduced female gene flow in the European flat oyster Ostrea edulis ArchiMer
Diaz Almela, F; Boudry, Pierre; Launey, Sophie; Bonhomme, Francois; Lapegue, Sylvie.
The geographical structure of 15 natural populations of the flat oyster (Ostrea edulis L.) was assessed by single-strand conformation polymorphism (SSCP) of a 313-base-pair (bp) fragment of the mitochondrial 12S-rRNA gene. Fourteen haplotypes were observed, with one being dominant in the Mediterranean samples and another one in the Atlantic populations. The geographically extreme populations sampled in Norway and the Black Sea appeared differentiated by exhibiting the dominance of a third group of haplotypes. The results were compared to available microsatellite data at five loci. The Atlantic/Mediterranean differentiation pattern was qualitatively the same with both types of markers, confirming an isolation-by-distance pattern. The average mitochondrial...

Between the cross and the sword: the crisis of the gene concept Genet. Mol. Biol.
El-Hani,Charbel Niño.
Challenges to the gene concept have shown the difficulty of preserving the classical molecular concept, according to which a gene is a stretch of DNA encoding a functional product (polypeptide or RNA). The main difficulties are related to the overlaying of the Mendelian idea of the gene as a ‘unit’: the interpretation of genes as structural and/or functional units in the genome is challenged by evidence showing the complexity and diversity of genomic organization. This paper discusses the difficulties faced by the classical molecular concept and addresses alternatives to it. Among the alternatives, it considers distinctions between different gene concepts, such as that between the ‘molecular’ and the ‘evolutionary’ gene, or between ‘gene-P’ (the gene as...

Plasmid patterns of efficient and inefficient strains of Bacillus thuringiensis against Spodoptera frugiperda (J.E. Smith) (Lepidoptera: Noctuidae) Neotropical Entomology
Fagundes,RBS; Picoli,EAT; Lana,UGP; Valicente,FH.
Bacillus thuringiensis harbors genes encoding Cry proteins found in chromosomes or plasmids of different sizes (4-150 Mb). Although the smaller plasmids are more abundant in B. thuringiensis, their specific function is unknown. As for the megaplasmids, their main recognized function is to harbor cry genes, although the sequencing of some of these plasmids indicates the occurrence of other important genes. This work used a new protocol for practical and rapid extraction of plasmid DNA in order to characterize the plasmid patterns of Brazilian strains belonging to Embrapa Milho e Sorgo research center B. thuringiensis bank. We tried to further assess the relationship of plasmid patterns with strains belonging to the same serovars and strains causing 100% and...

Associação de bioensaios e caracterização molecular para seleção de novos isolados de Bacillus thuringiensis efetivos contra Spodoptera frugiperda (J.E. Smith) (Lepidoptera: Noctuidae) Neotropical Entomology
Fatoretto,Júlio C.; Sena,Janete A.D.; Barreto,Marliton R.; Lemos,Manoel V.F.; Boiça Junior,Arlindo L..

Microarray analysis highlights immune response of Pacific oysters as a determinant of resistance to summer mortality ArchiMer
Fleury, Elodie; Huvet, Arnaud.
Summer mortality of Crassostrea gigas is the result of a complex interaction between oysters, their environment, and pathogens. A high heritability was estimated for resistance to summer mortality, which provided an opportunity to develop lines of oysters that were resistant (R) or susceptible (S) to summer mortality. Previous genome-wide expression profiling study of R and S oyster gonads highlighted reproduction and antioxidant defense as constitutive pathways that operate differentially between these two lines. Here, we show that signaling in innate immunity also operates differentially between these lines, and we hypothesize that this is at the main determinant of their difference in survival in the field. A reanalysis of our published microarray data...
